Description:
Trypsin Solutions
GIBCO?trypsin solutions are used widely for dissociation of tissues and cell monolayers. We can provide cell culture dissociating solutions that will perform consistently, batch after batch. The biochemical assays performed on Trypsin 1:250 determine both the trypsin-specific activity and the level of certain copurified enzymes that influence cell removal and viability. All trypsin solutions are prepared from porcine-parvovirus-tested and mycoplasma-tested materials. GIBCO?trypsin solutions are prepared under controlled conditions, using accelerated manufacturing procedures to minimize loss of enzyme activity from denaturation and autodegradation.

Raw Material Quality Control. Trypsin powdered raw material is subject to standard assays for solubility, loss of weight on drying, residue on ignition, and trypsin activity as outlined in the current edition of The United States Pharmacopeia, National Formulary (USP/NF).

Accepted values:
Activity. One USP trypsin unit is the activity causing a change in absorbance of 0.003/min in the presence of N-benzoyl-L-arginine ethyl ester hydrochloride at 25揅, pH 7.6. Trypsin (1:250) has an activity such that 1 g will digest (hydrolyze) 250 g of casein under the conditions of the USP test for protease activity of pancreatin.
Porcine Parvovirus. None detected.
Mycoplasma. None detected.
Amylase Activity. Acceptable.
Nucleotidase Activity. Acceptable.

Final Product Quality Control. GIBCO?enzyme preparations are tested to confirm the absence of bacterial and fungal contamination, employing protocols referenced in the current USP. The pH and osmolality of all batches are monitored using calibrated measuring equipment to verify conformance with product specifications.

In vitro bioassays are performed on the final product to verify the ability of the trypsin solutions to remove an adherent cell line from the substratum while preserving cellular integrity. The cell line selected for this assay is a human transformed cell line. The trypsin-dependent cell-removal times must be within a range established for each cell line. Following trypsinization, cells are replated and morphology is monitored after 18 to 24 h. Trypsin removal and replating are tested in parallel with a reference trypsin solution. Trypsin Replacement Enzymes
GIBCO甌rypLE?is a microbially produced alternative to animal trypsin. It is manufactured in a controlled fermentation process that is completely free of animal- and human-derived components. The process produces an exceptionally pure reagent, which is gentle on cells. Inactivation with trypsin inhibitors is not required. It can be stored refrigerated and is stable at room temperature for 6 months.

Gamma-Irradiated Liquid and Powdered Trypsin
Upon request, trypsin can be gamma-irradiated. We have validated the use of gamma-irradiation to inactivate certain porcine viruses which may be present in trypsin. Our studies indicate that an irradiation dose of 25 kGy will inactivate virus while preserving the biological performance in cell culture and the trypsin enzymatic activity.
