钙黄绿素 AM 是一种膜渗透性荧光酯酶底物，通常用于检测细胞活性和质膜完整性。作为一种近中性的底物，钙黄绿素 AM 可以自由、无创地扩散穿过质膜并渗透到活细胞的胞质溶胶中。一旦进入细胞内，代谢活跃细胞中的胞质酯酶会迅速将非荧光钙黄绿素 AM 底物水解成钙黄绿素产物，并由具有完整质膜的细胞保留。由于死细胞缺乏酯酶活性，因此仅标记活细胞。在蓝光激发下，钙黄绿素发出强烈的绿色荧光，与细胞酯酶的活性和样品中存在的活细胞数量成正比。与其他活细胞标记试剂（如 BCECF AM 或羧基荧光素二乙酸酯）相比，钙黄绿素 AM 荧光更亮、光稳定性更好，低细胞毒性有利于细胞追踪、细胞凋亡和细胞毒性的研究。
