Lys(Dde)是用于制造分支多肽,构建侧链环肽和进行Lys侧链荧光修饰的常用工具.
它的最大优点就是可以在保留多肽其它侧链保护基团的时候,单独被去除;也可以在去除其它保护基团的同时单独被保留.
Dde基团去除方法:用含2%水合肼的DMF(2% Hydrazine in DMF/DCM)反应30分钟
Method of selective removal of DDe with 2% hydrazine in DMF.
Resin
1. Place the peptidyl-Resin in a flask and treat with 2% hydrazine monhydrate in DMF (25ml/g). Stopper the flask and leave to stand at rt for 3 min.
2. Filter the resin and repeat the hydrazine treatment two more times. Wash the partially protected resin with DMF.
Peptide
1. Dissolve the Dde protected peptide with 2% hydrazine monhydrate in DCM(5mg/ml), stir at rt for 30 min.
2. Remove the DCM with vacuum.
3. Precipitate the deprotected peptide with cool ether and purified with HPLC.
